Use and maintenance of four incubators and precautions

The incubator is the main equipment for cultivating microorganisms and can be used for the cultivation and breeding of bacteria and cells. The principle is to artificially create an artificial environment in which the microorganisms and cells and bacteria grow and reproduce in the incubator, such as controlling a certain temperature, humidity, gas, and the like. The incubators currently used are mainly divided into four types: direct electrothermal incubators, water-blocking electrothermal incubators, biochemical incubators, and carbon dioxide incubators.

(1) Electrothermal and water-proof incubators The outer shells of electrothermal and water-isolated incubators are usually made of asbestos board or iron-painted paint. The inner layer of the water-proof incubator is a copper-colored water storage interlayer, and electrothermal culture. The interlayer of the box is made of insulating material such as asbestos or glass wool to enhance the heat preservation effect. The top of the incubator is provided with a thermometer and is automatically controlled by a temperature controller to make the temperature inside the box constant. The water-proof incubator is heated by means of an electric heating tube, and the electrothermal incubator is directly heated by a heating wire, and the air is convected to make the temperature inside the box uniform.
In the front side of the incubator, there are indicator lights and temperature adjustment knobs. When the power is turned on, the red indicator light is on, and the rotation is rotated to the required scale according to the required temperature. After the temperature is reached, the red indicator light is extinguished. It means that the required temperature has been reached in the box, and the temperature inside the box is automatically controlled by the temperature controller.
Use and maintenance:
(1) The culture in the box should not be over-squeezed to facilitate hot air convection. The items should be closed whenever the items are placed or removed to avoid temperature fluctuations.
(2) The electrothermal incubator should be placed in the tank with a container of water to maintain a certain humidity.
(3) The water-proof incubator should be carefully added with water and then energized. At the same time, the water level should be checked frequently and water should be added in time.
(4) The electric heating incubator should be properly unscrewed when used to facilitate the adjustment of the temperature inside the tank.

(2) Biochemical incubator This incubator is equipped with electric heating wire heating and compressor cooling. Therefore, it can be adapted to a wide range and can be kept at a constant temperature throughout the year, and thus is gradually popularized.
The incubator is similar to the electric heating incubator. Due to the installation of the compressor, the precautions for the maintenance of the refrigerator should also be observed. For example, keep the voltage stable, do not over-tilt, and clean the dust on the radiator in time.

(3) Carbon dioxide incubator The carbon dioxide incubator is improved on the basis of ordinary culture, mainly to add CO2 to meet the environment required for the cultivation of microorganisms. Mainly used for tissue culture and cultivation of some special microorganisms.
1. Installation and commissioning of CO2 incubator The front of the CO2 incubator has an operation panel with power switch, temperature regulator (manual and liquid crystal display), CO2 injection switch, CO2 adjustment knob, humidity adjustment knob. , temperature display panel, CO2 display panel and humidity display panel, carbon dioxide sample hole (for extracting the sample in the box to detect whether the CO2 in the box reaches the content displayed on the display panel) and alarm device (over temperature alarm light) .
(1) The incubator should be placed in a relatively stable location away from heat sources to prevent temperature fluctuations and microbial contamination.
(2) Before turning on the power, add a certain amount of distilled water to the incubator according to the instruction manual (it is best to add a certain amount of disinfectant to the added water, see the instruction manual) to avoid burning the machine.
(3) When the water is added to a certain amount, the alarm light is on, that is, the water is stopped, the power switch is turned on, and the heating is started, and the temperature controller is adjusted to the required temperature.
(4) When the temperature reaches the required temperature, the heating is automatically stopped. When the required temperature is exceeded, the over-temperature warning light is on and an alarm sounds.
(5) The CO2 used in the incubator can use liquid CO2 or gas, no matter which kind of CO2 is supplied, the pipe can not be too curved to ensure the smooth flow of gas. Generally, the CO2 cylinder is used, and the pressure control table can be connected.
(6) Zeroing of CO2 content After the temperature and humidity in the tank are stable (usually three days), turn the CO2 adjustment knob to adjust the number of the display panel to 0.00. Repeat the adjustment after 5 minutes. Show that the reading on the disc is stable. Turn on the CO2 injection switch (injection light is on), adjust the CO2 setting to the desired concentration so that the concentration reaches the set concentration (the injection lamp is extinguished) and maintain it for at least 10 minutes. Thereafter, the CO2 content in the tank is automatically adjusted by the CO2 controller.
(7) Adjusting the humidity of the CO2 incubator First press the humidity adjustment knob, then turn the humidity adjustment knob to the required culture humidity, and then adjust the humidity automatically by the humidity regulator.
2. Precautions for use (1) The incubator should be managed by a special person. Do not twist any switch and adjustment knob on the operation panel once it is fixed, so as not to affect the temperature inside the box, fluctuations in CO2 and humidity, and reduce the machine. Sensitivity.
(2) The water to be added must be distilled water or non-ionized water to prevent the mineral deposits from corroding in the water tank. Water must be changed every year. Always check if there is enough water in the box.
(3) The tank should be regularly disinfected with disinfectant, and the shelf can be taken out for cleaning and disinfection to prevent other microbial contamination, resulting in failure of the experiment.
(4) Regularly check the over-temperature safety device to prevent over-temperature. The method is to press the monitoring alarm button, turn the fixing screw until the over-temperature alarm device rings, and then turn off the over-temperature safety light.
(5) If carbon dioxide is not used for a long time, the CO2 switch should be turned off to prevent the CO2 regulator from malfunctioning.
(6) The CO2 used must be pure, otherwise reduce the sensitivity of the CO2 sensor and contaminate the CO2 filter unit.
(7) In the incubator without humidity control, in order to keep the CO2 in the tank stable, a water-filled container should be placed in the bottom of the tank.
There is also a three-gas incubator. This incubator is a new product that is further improved on the basis of a carbon dioxide incubator. The principle is the same as other incubators. It is characterized by not only adding CO2, but also adding nitrogen and oxygen, and all of them are controlled and regulated by various computers. It is mainly used for the reproduction and cultivation of some special microorganisms. It is generally not widely used.
